We tried adding DNA oligos to Dicty cells (with no transfection reagent), and observed levels of the associated protein increasing; this happened with oligos that would bind to the mRNA at the 5' UTR or coding region, and for 2 different genes.

Dear all,
Has anyone ever tried transient knock-down by RNA interference (siRNA)
with synthetic RNA oligos? Also have you used Lipofectamine as
transfection reagent for this purpose?
