We routinely perform electroporation in 0.2 cm cuvettes in an older BioRad Gene Pulser. The settings are 1.2 kV, 100 ohms and 3 uF. A time constant of 0.5 or 0.6 gives virtually 100% success; 0.4 is around 50% and less than 0.4 does not yield any
transformants. We always keep cells chilled for 10 after electroporation.
