Funny enough, we were just making plans for constructing an APEX2 that is targeted to the luminal side of the ER membrane. By having a signal sequence at the N-terminus, we hope that it is codon-optimized enough to be translated properly.

I’m also hoping to use APEX. I don’t think anyone has reported using it, though I expect codon usage will be important, as it was for BioID.
