Dear Dicty Researchers,

We have created a null mutant in Dicty that exhibits some developmental
defects. When we overexpress the knocked out gene from an extrachromosomal
plasmid under the act15 promoter some of the phenotypic defects are rescued,
but not the developmental defects. We would like to try rescuing by putting
the gene with its promoter back into the null mutant. Any advice on the
best way to do this so that the protein is expressed like the endogenous
gene (as much as possible) and what plasmid would be easiest to use is
appreciated.

Thanks,

Cynthia
