Back in the Firtel Lab we found that doing the sequencing reaction at a higher temperature helped

Subject: [DICTY] Difficulty in dictyostelium genome sequencing

Hello everybody,
I'm working in my project on Dictyostelium discoideum, in fact, we are facing a problem due to the richness of dicty genome with AT bases:  we aimed to do sequencing of some sequences we amplified by PCR. We sent our PCR products to GATC Biotech, and we failed to obtain a good sequencing for the most, probably due to the richness of dicty genome in AT bases.. I wonder if anybody has an experience in dicty sequencing? Is there a specific way to do sequencing for AT rich genome (complicated genome)?any advice or idea?
